چکیده
PC-1 is a nucleoside triphosphate pyrophosphohydrolase (NTPPPH) that generates pyrophosphate from the hydrolysis of ATP. PC-1 is the primary enzymatic generator of pyrophosphate in osteoblastic cells. The influence of PC-1 on tissue mineralization is complex (Figure 1). PC-1 activity increases extracellular levels of pyrophosphate. Pyrophosphate inhibits hydroxyapapatite crystal formation and propagation, yet PC-1 generated pyrophosphate also serves as an essential source of phosphate to enhance bone mineralization when it is hydrolyzed by the osteoblastic enzyme, alkaline phosphatase (AP). Additionally, while low pyrophosphate levels inhibit non-bone tissue calcification, dependent upon tissue composition, high pyrophosphate levels can lead to pathologic calcification of non-bone tissues. We previously showed that FGF2 specifically up-regulates PC-1 expression in MC3T3E1(C4) calvarial pre-osteoblasts, J Musculoskelet Neuronal Interact 2008; 8(4):318-320

FGF2 promotes Msx2 stimulated PC-1 expression via Frs2/MAPK signaling.
PC-1 is an enzymatic generator of pyrophosphate and a critical regulator of tissue mineralization. We previously showed that fibroblast growth factor-2 (FGF2) specifically induces PC-1 expression in calvarial pre-osteoblasts and that this occurs via a transcriptional mechanism involving Runx2. Because aberrant FGF signaling and Msx2 activity result in similar craniofacial skeletal defects and b...
متن کاملEffect of Oxidized Low Density Lipoprotein on the Expression of Runx2 and SPARC Genes in Vascular Smooth Muscle Cells
Background: Vascular calcification is an important stage in atherosclerosis. During this stage, vascular smooth muscle cells (VSMC) synthesize many osteogenic factors such as osteonectin (encoded by SPARC). Oxidative stress plays a critical role in atherosclerosis progression, and its accumulation in the vascular wall stimulates the development of atherosclerosis and vascular calcification. The...
متن کاملTranscriptional regulation of the human bone sialoprotein gene by fibroblast growth factor 2.
Fibroblast growth factor 2 (FGF2), a member of the FGF family, positively regulates bone formation and osteoblast differentiation. Bone sialoprotein (BSP) is highly expressed during early bone formation and may play a role in primary mineralization of bone. In the present study, FGF2 (10 ng/mL) was found to increase the levels of Runx2 and BSP mRNA at 3 and 12 h in human osteoblast-like Saos2 c...
متن کاملProlyl isomerase Pin1-mediated conformational change and subnuclear focal accumulation of Runx2 are crucial for fibroblast growth factor 2 (FGF2)-induced osteoblast differentiation.
Fibroblast growth factor 2 (FGF2) signaling plays a pivotal role in bone growth/differentiation through the activation of osteogenic master transcription factor Runx2, which is mediated by the ERK/MAPK-dependent phosphorylation and the p300-dependent acetylation of Runx2. In this study, we found that Pin1-dependent isomerization of Runx2 is the critical step for FGF2-induced Runx2 transactivati...
متن کاملBiphasic effects of FGF2 on odontoblast differentiation involve changes in the BMP and Wnt signaling pathways.
Odontoblast differentiation during physiological and reparative dentinogenesis is dependent upon multiple signaling molecules, including fibroblast growth factors (FGFs), bone morphogenetic proteins (BMPs) and Wingless/Integrated (Wnt) ligands. Recent studies in our laboratory showed that continuous exposure of primary dental pulp cultures to FGF2 exerted biphasic effects on the expression of m...
